药物AC-88对荷瘤鼠T细胞内TNF-α、lL-1β和IL-6 mRNA

EFFECTS OF AC-88 ON TNF-α、 IL-1β、 IL-6 mRNA CONTENTS AND PROLIFERATION OF T LYMPHOCYTE IN TUMOUR BEARING MICE

摘要: 目的:探讨AC-88抗肿瘤作用及可能机制。方法:以移植H22肝癌细胞的小鼠为体内模型,研究了AC-88的抑瘤作用;应用细胞培养和MTT法研究荷瘤小鼠T淋巴细胞的增殖功能;用RT-PCR法研究荷瘤小鼠T淋巴细胞TNF-α、IL-1β和IL-6 mRNA含量变化。结果:实验发现,AC-88的体内抑瘤率可高达76 %,而且AC-88能促进荷瘤鼠T淋巴细胞的增殖,能使T细胞中TNF-α、IL-1β、IL-6等细胞因子的mRNA含量增加2~4倍。 结论:AC-88有良好的抑瘤作用,还可以调节T淋巴细胞的功能,是一种具有较高开发价值的抗癌药物。

Abstract: Purpose: To investigate the antitumor effect and possible mechanism of AC-88. Methods: Tumor bearing mice were used as model in vivo to study the an titumor effect of AC-88. T cell proliferation was determined by using MTT method. The changes of TNF-α、 IL-1β、 IL-6 mRNA content in T cells were measured by RT-PCR. Results: The antitumor rate of AC-88 was 76 %. Treatmemt with AC-88 significantly enhanced T cell proliferation, and increased contents of TNF-α,IL-1β,IL-6 mRNA in T cells by 2-4 folds compared with that in control(P< 0.01) . Conclusions: AC-88 can inhibit the growth of tumor and regulate the T cell function in tumor bearing mice, which is worthy to be developed into an anticarcinogen.
